es marqueurs génétiques, particulièrement bien adaptés à ces problématiques car hypervariables, ont été mis au point récemment dans notre laboratoire en collaboration avec le laboratoire Génome et Populations de Montpellier (CNRS). Ils permettent désormais l'analyse approfondie et la discrimination fine de populations d'huîtres creuses Crassostrea gigas et de rechercher si certaines d'entre-elles n'auraient pas des caractéristiques intéressantes pour l'ostréiculture locale (croissance, résistance estivale, affinage ... ). La diffusion de telles populations auprès de la profession serait plus rapide car il s'agit de la même espèce. Le programme de sélection de l'huître plate a été initié en 1985 par des pathologistes de la Tremblade. La sélection ne portait que sur le seul critère de tolérance des huîtres plates au parasite Bonamia ostreae et correspondait plus à un outil pour étudier divers aspects de la parasitose qu'un un programme d'amélioration génétique visant la relance de la culture de l'huître plate. Dans le cadre du réseau REGEMO, ce programme a ensuite été repris par les généticiens de l'Ifremer : 1) rappel historique et définition d'un vocabulaire commun à l'ensemble des membres du réseau REGEMO. 2) Inventaire et bilan des opération menées ou en cours ( sur la dernière génération de sélection produite ne 1995, sur le matériel issu de cette génération et produit en 1997.... |

Dans le cadre du plan Etat-Région 1994-1998, un programme de conservatoire de souches a été initié au Laboratoire IFREMER de La Tremblade. Ce conservatoire a pour objectif d'acclimater différentes espèces ou souches d'huîtres creuses (genres Crassostrea et Saccostrea) afin d'acquérir des connaissances sur ces espèces et d'évaluer l'intérêt qu'elles pourraient présenter pour l'ostréiculture locale. L'importation d'espèces étrangères sur les côtes françaises est désormais strictement réglementée: la dissémination d'une nouvelle espèce peut se révéler néfaste d'un point de vue écologique et nuire aux productions existantes. De plus, ces huîtres peuvent être porteuses de pathogènes qui, s'il venaient à se répandre dans le milieu, pourraient toucher les stocks... |

Divers travaux, menés depuis 1984, montrent qu'il existe, chez l'huître creuse Crassotrea gigas, des cellules montrant un nombre anormal de chromosomes (2n = 19, 18 ou même 17 au lieu de 2n = 20). Le niveau d'aneuploïdie est déterminé par le décompte des chromosomes à partir de suspensions cellulaires de tissu branchial. Le pourcentage de cellules aneuploïdes est toujours significativement supérieur dans les "lots de queue", c'est-à-dire les huîtres présentant des croissances plus faibles, et peut atteindre plus de 30 %. The respective status of the Portuguese oyster, Crassostrea angulata, and the Pacific oyster, Crassostrea gigas, has long been a matter of controversy. Natural populations of the European flat oyster, Ostrea edulis, have been harvested since Roman times. This species is distributed along the European Atlantic coasts, from Norway to Morocco, as well as the Mediterranean Sea and Black sea coasts. Previous studies, based on allozymes, have concluded that isolation by distance is taking place in the Mediterranean sea and the Atlantic ocean. Because of the presumed non neutrality of these markers, another approach was made to examine the genetic differentiation of this species using nuclear microsatellite and mitochondrial markers, both considered as neutral, and to compare the results to those observed with allozymes. |

L'étude de la variabilité génétique de l' huître creuse Crassastrea gigas est un point essentiel dans la mise en place de programmes de gestion des ressources génétiques et de sélection chez cette espèce. C. gigas, originaire du Japon, a été introduite dans de nombreuses régions du monde et représente aujourd'hui plus de 95 % de la production mondiale d' huîtres. Crassostrea angulata and Crassostrea gigas, two cupped oysters of Asian origin (O'Foighil et al, 1998), have been introduced successively in Europe during the XVlth and the XXth century (Grizel et Héral, 1991). The present populations of C. angulata appear to be limited to the south of Spain and Portugal, whereas the distribution of C. gigas in Europe is rising because of the development of oyster farming. Crassostrea angulata and Crassostrea gigas, two cupped oyster taxa of Asian origin (O'Foighil et al, 1998), were successively introduced into Europe during the XVIth and the XXth century respectively (Héral and Grizel, 1991). To date, populations presumed to be C. angulata remain only in the south of Spain and Portugal, while the geographic range of C. gigas is expending, notably due to aquaculture. Two oyster species are cultivated on the French coasts: the Pacific cupped oyster (Crassostrea gigas), which has been introduced from Japan in the 1970's after the disappearance of the Portuguese oyster Crassostrea angulala, and the European flat oyster (Ostrea edulis). During the second half of the 1960's, the latter suffered successively from two diseases caused by protozoan parasites: marteiliosis (also called "maladie des Abers") and bonamiosis. In France, its production dropped from 20,000 tian in the 1960's to 2,000 today. The impact of these two pathogens as well as the exploitation of the remaining stocks show the importance of the conservation of this species, the only natural European species, whose population is more and more fragmented. |

Mangrove oysters include various species that are also present on different coasts. In the southern part of the Atlantic Ocean, Crassostrea rhizophorae has been observed along the southern coast of America and Crassostrea gasar along the western coast of Africa. Fifteen populations of these oysters were sampled from these two geographic areas. Their polymorphism was studied on the 16S mitochondrial fragment both by par sequencing and RFLP analysis. Some samples' caryotype were also analysed. Two haplotypes were identified: C. gasar was found in Africa, but also for the first time in South America, whereas C. rhizophorae was only found on the coasts of South America. |

Nowadays, PCR-based DNA markers allow the study of diversity and differentiation of marine organisms at different stages of their life cycle. Our study of European flat oyster (Ostrea edulis) populations has aimed to describe their spatial and temporal dynamics. First, we analyzed adults sampled over the natural range of the species.
